A lima bean core collection based on molecular markers Scientia Agricola

ABSTRACT Some germplasm collections have a high number of accessions, which makes it difficult to explore the genetic variability present in the germplasm bank due to the redundancy and the difficulty of detailed analysis of all conserved accessions. Therefore, our study aimed to analyze the genetic diversity of 153 lima bean (Phaseolus lunatus) accessions for the purpose of constructing a core collection. Eleven SSRs were used for this purpose. The 153 lima bean accessions can be represented by low redundancy using a minimum of 34 accessions, thus representing 22 % of the size of the entire germplasm bank. The objective of this study was to characterize the genetic variability of Anglo-Nubian goats using microsatellite markers. The study was conducted using herds from four municipalities of Central-Northern Piauí (Teresina, José de Freitas, Campo Maior, and Angical), where technical information is scarce. Seven markers suggested by FAO were used (ILSTS11, McM527, INRA23, ETH10, OarfCB304, OarfCB48, and MAF209). The samples were genotyped using a 7% polyacrylamide gel. Caffeine inheritance in interspecific hybrids of Coffea arabica x Coffea canephora (Gentianales, Rubiaceae) Genet. Mol. Biol.

Caffeine inheritance was investigated in F2 and BC1F1 generations between Coffea arabica var. Bourbon Vermelho (BV) and Coffea canephora var. Robusta 4x (R4x). The caffeine content of seeds and leaves was determined during 2004 and 2005. Microsatellite loci-markers were used to deduce the meiotic pattern of chromosome pairing of tetraploid interspecific hybrids. Genetic analysis indicated that caffeine content in seeds was quantitatively inherited and controlled by genes with additive effects. Genetic diversity of 60 Hevea genotypes, consisting of Asiatic, Amazonian, African and IAC clones, and pertaining to the genetic breeding program of the Agronomic Institute (IAC), Brazil, was estimated. Genetic diversity among Brazilian soybean cultivars based on SSR loci and pedigree data BABT

In this study, simple sequence repeats (SSR) loci and pedigree data were used to investigate the genetic relationship in a group of 168 Brazilian soybean cultivars. Eighteen SSR loci produced an average of 5.06 alleles and a mean gene diversity of 0.58 for the cultivars studied. Genetic distance (GD) was determined using the modified Roger's Wright distance, and a final dendrogram was in agreement with the cultivar pedigree. A distance matrix based on the coefficient of parentage scores was also generated for the cultivars, which ranged from 0 to 1, with a mean of 0.18, whereas SSR-based genetic similarity (1- GD) ranged from 0.01 to 0.90, with a mean of 0.25. The pigeonpea [Cajanus cajan (L) Millspaugh] is one of the most important perennial legume crops utilized in the food, fodder, soil conservation, crop-livestock integrated systems, reclaiming of degraded pastures and symbiotic nitrogen fixation. Microsatellite markers were used to estimate the genetic diversity of 77 pigeonpea genotypes selected from the germplasm collections at Embrapa Cattle-Southeast and, to evaluate their transferability to Phaseolus vulgaris and Vigna unguiculata species. The RADP (Random amplified polymorphic DNA) technique was used to detect tissue-culture-induced variations in sugarcane. Plants of the Brazilian variety RB83-5486 propagated via rhizomes and via meristem cultures were studied. The polymorphism rate for 98 RAPD loci was 6.93% when the plants derived from meristems. Besides, in order to evaluate the influence of the number of subcultures on the generation of somaclonal variation, field-grown RB83-5486 plants derived from 10 meristems were studied after five subcultivations. The ''cagaita tree'' (Eugenia dysenterica) is a plant found widespread in the Brazilian Cerrado. Its fruit is used for popular consumption and for industrial purposes. This study opens a new perspective for the generation of population genetic data and parameters estimates for devising sound collection and conservation procedures for Eugenia dysenterica. This study was carried out to assess the genetic variability of ten "cagaita" tree (Eugenia dysenterica) populations in Southeastern Goiás. Fifty-four randomly amplified polymorphic DNA (RAPD) loci were used to characterize the population genetic variability, using the analysis of molecular variance (AMOVA). A phiST value of 0.2703 was obtained, showing that 27.03% and 72.97% of the genetic variability is present among and within populations, respectively. We used eight microsatellite loci to estimate genetic structure and investigate within and between population genetic variation in eleven Brazilian wild rice (Oryza glumaepatula) populations. Traditional growers of the Vale do Ribeira, São Paulo State, grow and make use of several D. alata landraces. This study assessed the genetic diversity of 16 landraces, using isozymatic and morphological markers and comparing them with 19 commercial varieties of D. alata. O objetivo do trabalho foi relacionar a diversidade genética medida a partir de três diferentes marcadores moleculares, com a variação genética quantitativa de caracteres poligênicos, estimada em ensaio de progênies, sob condições controladas. As progênies, oriundas de dez subpopulações naturais de cagaiteira do sudeste de Goiás, foram avaliadas em experimento em blocos completos casualizados, com quatro repetições e uma planta por parcela. Foram estimadas a herdabilidade ao nível de média de progênies (h mi ²) e o coeficiente de variação genética (CVgi) de cada subpopulação para a altura da planta e o diâmetro do fuste, por quatro anos, bem como para as respectivas taxas de crescimento. Similaridade genética entre raças bovinas brasileiras PAB

A similaridade genética entre animais de duas raças bovinas brasileiras (Crioulo Lageano e Junqueira) foi estimada pela análise de polimorfismo de DNA amplificado ao acaso (RAPD), tendo como referência (outgroups) animais de raças comerciais das espécies Bos taurus e B. indicus. Estas duas raças possuem grande similaridade fenotípica, sugerindo uma origem genética comum. Uma matriz de similaridade genética baseada em polimorfismo de DNA foi obtida e representada graficamente por um dendrograma, definido após processo estatístico de reamostragem bootstrap.
